1.22.3 Step 2 - Gibson Assembly: Select Backbone

• In this stage you can choose to select a backbone and then import it into the wizard.

Uploading a Backbone

• Gibson Assembly can be circular or linear. For circular Gibson Assembly you will require a backbone. The "Yes" option is selected by default.

• You can upload a vector sequence by either drag and drop from the Materials box, choose a file from your computer or paste/type a sequence (Figure 1.22.3.1). For linear Gibson Assembly, select "No" and then click "Next."

• Once you selected the backbone, select the "Linearise method": "By Digest", "By PCR", or "By Sequence Position" (Figure 1.22.3.2).
